We are excited to announce that all reaction buffers are now BSA-free. NEB began switching our BSA-containing reaction buffers in April 2021 to buffers containing Recombinant Albumin (rAlbumin) for restriction enzymes and some DNA modifying enzymes. Find more details at www.neb.com/BSA-free.
EnGen® Sau Cas9 is a programmable DNA endonuclease guided by a single guide RNA (gRNA). Targeting requires gRNA complementarity to the target site as well as a 5′NNGRRT protospacer adjacent motif (PAM) on the DNA strand opposite the target sequence. Cleavage by EnGen® Sau Cas9 occurs ~3 bases upstream of the PAM and leaves blunt ends.
Longer 5′ NNGRRT 3′ PAM sequence allows for higher specificity
Two nuclear localization signals for improved transport into the nucleus
High concentration liquid format option can be used for microinjection, electroporation and lipofection
Compatible with the EnGen® Mutation Detection Kit
Need guide RNA? NEB recommends the Horizon™ CRISPR Design Tool for designing and ordering synthetic guide RNA for EnGen Sau Cas9
EnGen® Sau Cas9 from Staphylococcus aureus is an RNA-guided DNA endonuclease that catalyzes site-specific cleavage of double-stranded DNA. Targeting requires a ~100 nucleotide guide RNA (gRNA) complementary to the target site, as well as a 5′ NNGRRT 3′ (N = any, R = A or G) protospacer adjacent motif (PAM) on the DNA strand opposite the target sequence. DNA cleavage by EnGen Sau Cas9 occurs ~3 bases 5′ of the PAM. EnGen Sau Cas9 has Simian virus 40 (SV40) T antigen nuclear localization signals (NLS) at both the N- and C-termini of the protein.
